    What is a Sickling Test Test?

    The Sickling test is a diagnostic procedure used to detect the presence of sickle hemoglobin (HbS) in the blood, aiding in the diagnosis of sickle cell disease (SCD). Sickle cell disease is an inherited condition resulting from a mutation in the beta-globin gene, where valine replaces glutamic acid at the sixth position, causing red blood cells to form an abnormal crescent or "sickle" shape. These abnormally shaped cells have difficulty passing through small blood vessels, often leading to vaso-occlusive crises, which can result in severe pain and damage to organs and tissues. The Sickling test uses the solubility method, wherein sodium dithionite is added to a phosphate buffer containing the blood sample, leading to the crystallization of HbS, which results in turbidity. This turbidity is compared to control samples to confirm the presence of HbS. Although the test is both simple and economical, it has certain limitations. It may yield false negatives in neonates due to the high levels of hemoglobin F, or in individuals with co-inherited alpha-thalassemia or severe anemia. Moreover, the test cannot distinguish between sickle cell trait (SCT) and sickle cell disease (SCD), and false positives may occur in cases of erythrocytosis or significant leukocytosis.
